Virus-Specific T cells: Promising Adoptive T cell Therapy against Infectious Diseases following Hematopoietic Stem Cell Transplantation
نویسندگان
چکیده
Hematopoietic stem cell transplantation (HSCT) is a life-saving therapy for various hematologic disorders. Due to the bone marrow suppression and its long recovery period, secondary infections, like cytomegalovirus (CMV), Epstein-Bar virus (EBV), adenovirus (AdV), are leading causes of morbidity mortality in HSCT cases. Drug resistance antiviral pharmacotherapies makes researchers develop adoptive T therapies virus-specific therapy. These studies have faced major challenges such as finding most effective expansion methods, isolating expected subtype, defining functionality end-cell population, product quality control, clinical complications after injection. This review discusses viral infections HSCT, cells characteristics during chronic infection, application (VSTs) refractory standard methods producing VSTs their limitation, experiences on VSTs, focusing outcomes side effects that can be helpful decision-making patients further researches.

The introduction and evolution of hematopoietic stem cell transplantation (HSCT) could be traced back to 1950s, to the studies on interactions among irradiation, covering spleen and bone marrow from it and injection of bone marrow cells. Today, HSCT is considered a well-established, effective and promising means of therapy for various malignant and non-malignant medical conditions, both in chil...
متن کاملHematopoietic Stem Cell Transplantation for Thalassemia
Thalassemia is an autosomal recessive disorder associated with defective synthesis of the α- or β-chain of hemoglobin. For β-thalassemia major patients, therapeutic options are either monthly red cell transfusions and chelation therapy or allogeneic stem cell transplant. Stem cell transplant is the only curative approach and success is inversely correlated with the degree of iron overload and h...
متن کاملT-Cell Therapy: Options for Infectious Diseases
The emergence of drug-resistant tuberculosis is challenging tuberculosis control worldwide. In the absence of an effective vaccine to prevent primary infection with Mycobacterium tuberculosis and tuberculosis disease, host-directed therapies may offer therapeutic options, particularly for patients with multidrug-resistant and extensively drug-resistant tuberculosis where prognosis is often limi...
متن کاملPrimary allogeneic T-cell responses against mantle cell lymphoma antigen-presenting cells for adoptive immunotherapy after stem cell transplantation.
PURPOSE In patients treated with allogeneic stem cell transplantation for advanced mantle cell lymphoma (MCL), complete sustained remissions have been observed illustrating susceptibility of MCL cells to a graft-versus-lymphoma effect. To potentiate this graft-versus-lymphoma effect, adoptive transfer of in vitro selected MCL-specific CTL can be an attractive approach. The lack of expression of...
متن کاملGeneration of human memory stem T cells after haploidentical T-replete hematopoietic stem cell transplantation.
Memory stem T cells (TSCM) have been proposed as key determinants of immunologic memory. However, their exact contribution to a mounting immune response, as well as the mechanisms and timing of their in vivo generation, are poorly understood. We longitudinally tracked TSCM dynamics in patients undergoing haploidentical hematopoietic stem cell transplantation (HSCT), thereby providing novel hint...
